劉廷析先生(1967.12.22-2011.07.16),貴州省貴陽市人,研究員、博士生導(dǎo)師。中國科學(xué)院上海生命科學(xué)研究院健康科學(xué)研究所/上海交通大學(xué)醫(yī)學(xué)院健康科學(xué)研究所所長助理,發(fā)育與疾病實(shí)驗(yàn)室研究組長,研究員,博士生導(dǎo)師。上海交通大學(xué)醫(yī)學(xué)院瑞金醫(yī)院上海血液學(xué)研究所醫(yī)學(xué)基因組學(xué)國家重點(diǎn)實(shí)驗(yàn)室研究組長,教授,博士生導(dǎo)師。上海高校模式生物E—研究院特聘教授,貴陽省貴陽醫(yī)學(xué)院客座教授,上海交通大學(xué)特聘教授。美國血液學(xué)會(huì)會(huì)員。
1996獲武漢同濟(jì)醫(yī)科大學(xué)血液學(xué)碩士學(xué)位,1999年獲上海第二醫(yī)科大學(xué)血液學(xué)博士學(xué)位。美國哈佛大學(xué)醫(yī)學(xué)院Dana-Farber癌癥研究所博士后。
2004年獲中國科學(xué)院“百人計(jì)劃”;2005年獲“國家杰出青年科學(xué)基金”;2006年獲上海浦江人才計(jì)劃,并承擔(dān)中國科學(xué)院知識(shí)創(chuàng)新工程重要方向項(xiàng)目;2007年作為首席科學(xué)家承擔(dān)國家科技部973重大科學(xué)研究計(jì)劃項(xiàng)目;2008年承擔(dān)國家自然科學(xué)基金重點(diǎn)項(xiàng)目。2009年上海市科委優(yōu)秀學(xué)科帶頭人。2011年參加科技部973計(jì)劃 “造血干細(xì)胞維持、衰老與再生的調(diào)控機(jī)制研究”(2011CB964800),2011年入選中國科學(xué)院戰(zhàn)略性先導(dǎo)科技專項(xiàng)研究團(tuán)隊(duì),承擔(dān)子課題“使用前向遺傳學(xué)策略識(shí)別中胚層和造血干細(xì)胞自我更新的信號傳導(dǎo)網(wǎng)絡(luò)”。
2007年獲“新世紀(jì)百千萬人才工程國家級人選”及“2007年度明治乳業(yè)生命科學(xué)獎(jiǎng)”。2008年獲得“第四屆上海青年科技英才提名獎(jiǎng)” “2008健康科學(xué)研究所優(yōu)秀研究員”及“政府特殊津貼”。2010年度當(dāng)選上海交通大學(xué)首屆“吾愛吾師”十佳導(dǎo)師。2010年獲“談家楨生命科學(xué)創(chuàng)新獎(jiǎng)”。
主要成果: 主要使用斑馬魚發(fā)育-疾病模式生物體,進(jìn)行人類造血系統(tǒng)發(fā)育和疾病的分子遺傳學(xué)及發(fā)育生物學(xué)研究和轉(zhuǎn)化型醫(yī)學(xué)研究。2000年,闡明急性早幼粒細(xì)胞白血病維甲酸誘導(dǎo)分化的分子網(wǎng)絡(luò)調(diào)控機(jī)制(Blood, 2000)。2002年,澄清人類5號染色體長臂5q31和5q32-34上兩個(gè)白血病關(guān)鍵缺失區(qū)4.5億年的進(jìn)化規(guī)律(PNAS, 2002)。2003年,通過使用斑馬魚發(fā)育-疾病模式生物體,揭示人類遺傳性白血病Fanconi貧血病人發(fā)育缺陷的分子遺傳學(xué)機(jī)制(Developmental Cell, 2003)。2007年,發(fā)現(xiàn)人類5號染色體長臂5q31白血病關(guān)鍵缺失區(qū)內(nèi)白血病腫瘤抑制基因及其表觀遺傳學(xué)失活機(jī)制(Nature Medicine, 2007)。在此工作基礎(chǔ)上,2010年發(fā)現(xiàn)一個(gè)與白血病發(fā)生相關(guān)的分子調(diào)控路徑:PTEN-C/EBPalpha- CTNNA1,該白血病干細(xì)胞腫瘤抑制軸的發(fā)現(xiàn)也為白血病干細(xì)胞的靶向治療(特別是CTNNA1表達(dá)低的患者)提供了重要線索(Blood, 2010)。2008年,率先建立在動(dòng)物活體內(nèi)實(shí)時(shí)、動(dòng)態(tài)示蹤巨噬細(xì)胞遷移的斑馬魚模型(Journal of Immunology, 2008)。同年,使用斑馬魚作為研究模型,通過深入的生物信息學(xué)分析,在全基因組規(guī)模上確定了62個(gè)含有Set結(jié)構(gòu)域、具有潛在組蛋白甲基化酶活性基因的時(shí)空表達(dá)譜式和進(jìn)化規(guī)律(PLoS One, 2008);在此基礎(chǔ)上,2010年闡述器官左-右不對稱發(fā)育起源新機(jī)制,該研究首次提示組蛋白甲基化介導(dǎo)的表觀遺傳學(xué)機(jī)制在背側(cè)組織者和左右不對稱軸建立中起重要作用(PNAS, 2010)。2009年,解析微小RNA基因miR-144選擇性調(diào)節(jié)紅細(xì)胞血紅蛋白合成的環(huán)路機(jī)制(Blood,2009),并首次建立斑馬魚條件性knockdown的理論和技術(shù)體系(PLoS ONE, 2009)。2011年建立了造血組織特異性轉(zhuǎn)基因斑馬魚模型,在胚胎造血干細(xì)胞發(fā)育研究和血液疾病發(fā)生研究間架設(shè)了“橋梁”,為之后的藥物和治療靶點(diǎn)的篩選提供了平臺(tái)和基礎(chǔ)(Blood,2011)。實(shí)驗(yàn)室自2005年成立,近五年在國際學(xué)術(shù)期刊上發(fā)表研究論文19篇,總計(jì)IF>145。
